Abstrakt

This study aims at correlating ice crystal structures which form during the slow freezing of human red blood cells, to the mechanism of slow-freezing injury of cells. The process of directional solidification of suspensions with human red blood cells was observed for various cooling rates. Two kinds of solutions for suspension were used: physiological saline and physiological saline with 20 % v/v glycerol, a common cryoprotectant. The characteristics of ice crystals and the natures of interaction between ice crystals and red blood cells were morphologically investigated. It was found that the patterns of interaction between the ice crystals and the cells are remarkably different between the two solutions. Effects of mechanical protection of glycerol were discussed, and classification of the patterns of interaction was proposed.
